Metabolomics is an omics method to analyze all metabolites of a certain biological component or cell quantitatively and qualitatively under a specific physiological or pathological condition. It can be used for early disease diagnosis, drug target discovery, and disease mechanism research. Metabolomics mainly includes untargeted metabolomics and targeted metabolomics.

1. Untargeted metabolomics: In a set of experimental and control samples, it detects metabolites with statistically significant changes in abundance, identifies their structure, and reveals the correlation between the discovered metabolites and physiological or pathological conditions.

2. Targeted metabolomics: The research target is a certain type of compound or a compound related to a certain metabolic pathway. The target ion pair can be selected for high-sensitivity quantitative analysis using MRM mode.

S-adenosylmethionine (SAM) is a biochemical methyl donor that is catalyzed by methionine adenosyltransferase with ATP involved. SAM demethylates under the action of methyltransferase to form S-adenosylhomocysteine (SAH). Finally, SAH is hydrolyzed to remove adenosine and converted to homocysteine.

SAM and SAH are widely present in various organisms. As important physiologically active substances, they participate in the methylation of body metabolism and are closely related to many key bodily functions including gene regulation, hormone activity, nerve conduction development and detoxification. They show a wide range of therapeutic effects, have good efficacy in the treatment of liver dysfunction, depression, arthritis, and other diseases, and are important indicators of Alzheimer’s disease (AD), Parkinson’s disease (PD), systemic lupus erythematosus, cancer and many other diseases.

In a collaboration with a research group in the Department of Molecular Pharmacology, Albert Einstein College of Medicine, we provided services to quantitatively analyze SAM and SAH levels in plasma and liver samples. The SAM methyl cofactor and its SAH methyltransferase product were analyzed in plasma. In brief, plasma samples were spiked with an internal standard solution in 0.1% formic acid and then treated with 70% perchloric acid. Following acid extraction, supernatants were analyzed by HPLC-tMS in the multiple reaction monitoring (MRM) setting. For liver SAM and SAH, HPLC was used to analyze acid extracted rat liver samples. Both SAH and SAM were characterized by a maximum absorption at 256 nm and eluted at 11.39 and 12.76 min, respectively; such detection was linear across a 5-140 pmol range.
